How to Pre-Portion Microdoses Into Capsules for Consistent Dosing
By Microdosing College Editorial·Last updated August 1, 2026·7 min read
If you are learning how to pre-portion microdoses in capsules, the single most important thing to understand first is why the effort matters: psilocybin content varies significantly between individual truffles in the same bag, between bags from the same harvest, and between batches from different suppliers. A preregistered field study by Marschall et al. (2022) explicitly documented this "dose and potency variability of truffle batches" as a core methodological limitation — meaning even researchers working under controlled conditions cannot assume each piece of truffle delivers the same amount of active compound. Pre-portioning into capsules is not about perfection; it is about making your doses more consistent than tearing off a piece by eye, one day at a time.
This article is part of the microdosing psilocybin pillar. Before you begin, run through our to check for contraindications — capsules do not change who should or should not microdose; they only change a dose is prepared. Nothing in this guide is a recommendation to take any substance.
Fresh truffles are approximately 85–90% water by weight. A 2 g fresh microdose becomes roughly 0.2–0.3 g dry — a tenfold reduction. If you try to weigh and capsule fresh material, the continuing moisture loss will mean your capsules are heavier on day one and lighter by day fourteen, which defeats the purpose entirely.
How to dry truffles for capsules:
Slice truffles as thinly as practical (2–3 mm). Greater surface area speeds drying and reduces the time at which moulds can develop.
Use a food dehydrator set to 35–40 °C (95–104 °F) for 4–8 hours. Temperatures above 50 °C begin to degrade psilocybin (Marschall et al., 2022), so keep the heat low.
If you have no dehydrator, lay slices on a clean wire rack in a well-ventilated room, away from light, for 24–72 hours. A small desk fan helps.
The finished product should snap cleanly like a cracker. If it bends, it still contains too much moisture and will cause capsules to clump.
Drying alone does not solve the variability problem, because psilocybin is not distributed evenly across a truffle. The outer skin and certain denser regions can be higher in active compounds than the pale interior. Homogenising — grinding the entire dried batch into a fine, uniform powder — is the most important step in the whole workflow.
Use a clean coffee grinder or spice grinder dedicated only to this purpose. Residual coffee oils affect taste and introduce unknown variables.
Grind in short pulses (3–4 seconds each) rather than one long run, which can generate heat.
Aim for a consistent fine-powder texture, similar to flour. Coarse chunks will fill capsules unevenly.
Powder the entire batch at once. Grinding in portions and mixing them is less effective than a single combined grind.
Once you have a uniform powder, the concentration of active compounds per gram of material is as consistent as it can be, given natural biological variation. This is the underlying logic: you cannot eliminate potency variability entirely, but homogenisation distributes it as evenly as possible across every capsule.
Step 3 — Weigh doses precisely
This is where accuracy lives. A kitchen scale that rounds to the nearest gram is not adequate. You need a jewellery or milligram scale accurate to 0.01 g (10 mg). Widely available online for under €20, these scales make a meaningful practical difference: a 0.05 g rounding error on a 0.1 g target dose is a 50% error.
For how much constitutes a microdose in grams, the standard range for dried psilocybin material is typically 0.05–0.3 g dry weight, with many people starting at 0.1 g. Our microdosing dosage guide covers how to find your personal starting point — but for first-time batches, conservative is always safer.
Weighing workflow:
Tare (zero) the scale with an empty capsule half in the weighing pan.
Add powder with a small scoop or spatula until you reach your target weight.
Note the weight of each capsule in a log — this builds a record of your batch consistency.
If a capsule is slightly over, remove a small amount; if under, add a pinch.
For a typical 0.1 g target dose and a standard size-0 capsule, you will find the capsule fills comfortably without needing to tamp or compress, which can crack capsule walls.
Step 4 — Fill and close the capsules
Vegetarian (HPMC) or gelatine capsules in size 0 or 00 are readily available from supplement suppliers. Size 0 holds approximately 500 mg of a dense powder; truffle powder is lighter, so it holds less by weight — which is fine, because your microdose is well under that ceiling.
Use a capsule filling tray if you are making more than 10–15 capsules. Trays hold the lower halves upright while you fill them and dramatically speed up the process.
Fill the bottom half, then press the top half on firmly until you feel a click.
Do not lick capsules to seal them. Saliva introduces moisture and can affect storage life.
Wipe the outside of each capsule with a dry cloth to remove any powder residue before storing.
Step 5 — Label and store
Labelling is a harm-reduction necessity, not an optional nicety. Unlabelled capsules containing an active substance should never be stored where they could be mistaken for supplements, vitamins, or medication by household members who did not prepare them.
Each storage container should show:
Substance and batch date (e.g., "PT — dried truffle powder, batch 2026-06-01")
Target dose per capsule in grams (e.g., "0.10 g")
Number of capsules in the container
A clear warning that these are not food supplements
Storage conditions: light and heat degrade psilocybin. Store capsules in an airtight, opaque container (a dark glass jar with a silica gel desiccant packet works well) in a cool, dark location. A refrigerator is suitable if the container is truly airtight; condensation from repeated opening is the main risk. Under these conditions, dried capsules typically remain stable for several months, though no peer-reviewed data on precise capsule shelf life for truffle powder specifically are available.
Consistency, not certainty: setting realistic expectations
Pre-portioned capsules meaningfully reduce preparation variability — the error introduced by eyeballing doses or working with moist material. They do not eliminate the underlying biological variability in truffle potency, which remains a documented limitation even in research settings (Marschall et al., 2022).
This is why tracking your experience remains important alongside capsule preparation. As described in microdosing journal: what to track, a simple daily log of dose, mood, energy, sleep, and any unwanted effects lets you spot patterns that scale readouts alone cannot show. If you notice that one batch feels noticeably stronger or weaker than the previous one at the same weight, that is normal biological variation — adjust your dose conservatively rather than making large changes.
For protocol options — including whether to microdose on a Fadiman schedule or an alternative — see /protocols. If at any point you are unsure whether what you are experiencing is a side effect, an interaction, or simply a stronger-than-expected batch, our signs a microdose is too low or too high guide describes the key indicators. Systematic adverse effects (persistent nausea, elevated anxiety, cardiovascular symptoms) have been documented even at sub-perceptual doses in some individuals (Yerubandi et al., 2024); do not ignore them.
Why do capsules give more consistent microdoses than tearing off a piece of truffle?+
Fresh truffle pieces vary in psilocybin concentration across the same batch. Drying and grinding the entire batch into a uniform powder distributes potency as evenly as possible; weighing each capsule on a milligram scale then gives each dose the same dry weight, reducing preparation error significantly.
What capsule size should I use for microdosing?+
Size 0 vegetarian (HPMC) or gelatine capsules work well for typical microdoses of 0.05–0.3 g of dried truffle powder. Size 00 is larger and can feel unwieldy for small doses. Both are widely available from supplement suppliers.
Does drying truffles destroy psilocybin?+
High heat does degrade psilocybin. Research contexts caution against temperatures above 50 °C. Drying at 35–40 °C in a food dehydrator, or air-drying on a rack, preserves the active compounds while removing moisture that would cause dose weights to drift over time (Marschall et al., 2022).
How long can pre-filled microdose capsules be stored?+
Properly dried and sealed capsules stored in an airtight, opaque container in a cool dark place can remain stable for several months, but peer-reviewed shelf-life data specific to truffle-powder capsules are not available. Silica gel desiccant packets reduce moisture-related degradation.
Is capsule preparation legal in the Netherlands?+
In the Netherlands, psilocybin truffles (sclerotia) remain legal to sell and possess for adults. Preparing them into capsules for personal use as part of an informed microdosing practice is consistent with that legal status — however, this does not constitute legal advice, and rules differ outside the Netherlands.
